So I entered in the building, talked with a girl entered in the bar and a girl started shooting me. I run away went back after a little without guns this time (maybe this is the reason why they shoot at me?) but the door is closed. Was there anything really really important in the bar ? Have I damaged the stoyryline forever ?

They attacked you because of the guns (read the sign at the entrance).

The door was closed because of the bug in the kill counting code which is fixed in patch 1.0.1.

You haven't broken the storyline, you can get back into the bar through the alley with the crazy bum, or you can use the 'opensesame' cheat on the door to open it up again.
